In case people weren't aware, NX Dundee have recently started night bus services, so perhaps if that is a success they may have a go in the West Midlands again?

The best route to trial a Night Bus would in my eyes would be
Walsall
Stourbridge
Sutton Coldfeild

Best Route to Trial a 24hr Service would be
900 as it is close enough to 24HR

Most major cities have a night bus network why dont we, and why not just trial a flat fare £1.50. When i go to London Leeds Liverpool Manchester its suprising at how many people i see on them, and then you come home to Birmingham and all buses have left at Midnight.

I'm sure a nighttime 3 (following 503 route, not 503A+W as now) would go down well with the night shift workers on the Stafford Road corridor (and later i54). There are a lot of overnight workers at Freshway, Goodrich, and the other places. That's a <15 minute route. I wouldn't take it down Waterloo Road or around Wobaston though at that time of night (though Asda is 24 hours).
My local's the 3 and 63.
